During my PhD, I performed toxicity studies on living pancreatic cells that produce insulin in the presence of glucose. The cells were placed in tissue chip devices for a continuous exposure to the fluidic flow of nutrients. Series of stimuli would then be exposed and using live-cell imaging microscopy technique, we would be able to monitor cellβs ability to respond to the challenge. Here is a cool movie that I made (exposure to the different stimuli is animated on the top of the panel).
What you see is a cell (in green heat-map) on left firing robust calcium signals (surrogate for insulin release) when exposed to high glucose + toxic agent compared to the cell treated with basal glucose + no toxic agent on the right. Cellular death is visualized in magenta in both cells.
Here is a visualization of data to highlight strategies for reducing variability in data generation. Data replication of 20+ is plotted and animated on the same graph.
